1、YB1表達(dá)對乳腺癌細(xì)胞遷移和侵襲能力的影響作者：楊靖許文林李娟弓晉靈吳曉君陳巧云王法春【摘要】　　目的：觀察Y盒結(jié)合蛋白1（YB1）對腫瘤細(xì)胞遷移和侵襲能力的影響。方法：通過脂質(zhì)體介導(dǎo)的方法將YB1shRNA重組載體轉(zhuǎn)染入人類乳腺癌MCF7細(xì)胞，經(jīng)G418篩選及克隆化培養(yǎng)后獲得穩(wěn)定轉(zhuǎn)染株。運(yùn)用RTPCR和蛋白質(zhì)印跡方法檢測MCF7細(xì)胞中YB1mRNA和YB1蛋白的表達(dá)水平，運(yùn)用劃痕實(shí)驗(yàn)和Transwell小室法檢測細(xì)胞運(yùn)動、遷移和侵襲能力的變化，同時(shí)運(yùn)用RTPCR和蛋白質(zhì)印跡方法檢測細(xì)胞中MMP2的表達(dá)水平，并用明膠酶譜法檢測MMP2蛋白的活性。結(jié)果：穩(wěn)定
