6、onandcollagenproductionbyVitE.【Keyiasisjaponica;livercirrhosis;vitaminE;oxidativestress;hepaticstellatecell(HSC);collagentypeI【摘要】目的:探討維生素E(VitE)抗小鼠日本血吸蟲病肝纖維化作用及其機(jī)制.方法:用日本血吸蟲尾蚴皮膚敷貼法感染小鼠,構(gòu)建日本血吸蟲病肝纖維化模型,以肝纖維化達(dá)Ⅱ級(jí)或Ⅱ級(jí)以上作為開始VitE治療標(biāo)志.實(shí)驗(yàn)分5組:正常對(duì)照組、模型組及VitE高、中、低劑量組(每日150,50,5mg
7、/kg),8RNA表達(dá).結(jié)果:VitE降低模型組肝臟MDA含量[(5.00±0.31)μmol/gvs(11.66±1.84)μmol/g,P<0.05],提高SOD活性[(249.84±26.22)μkat/gvs(120.11±42.61)μkat/g,P<0.05];減少αSMA陽(yáng)性表達(dá)[(0.41±0.02)vs(0.68±0.02),P<0.05];降低肝臟膠原含量[(0.23±0.01)vs(0.60±0.11),P<0.05]和α1(I)型前膠原基因表達(dá)[(0.28±0.01)vs(0.85±
8、0.15),P<0.05].結(jié)論:VitE具有抗小鼠日本血吸蟲病肝纖維化作用,其機(jī)制與抗脂質(zhì)過(guò)氧化作用、抑制HSC活化和增殖、降低α1(I)型前膠原基因表達(dá)和膠原合成有關(guān).【關(guān)鍵詞】血吸蟲病,日本;肝硬化;維生素E;氧化性應(yīng)激;肝星狀細(xì)胞;膠
