1、丹參酮IIA增效大鼠骨髓源性內(nèi)皮祖細胞VEGF、SDF-1表達楊彥陳慶偉曹廣煜李桂瓊(重慶醫(yī)科大學(xué)附屬第二醫(yī)院老年心血管病科,重慶400010)【摘要】目的研究在丹參酮IIA的輔助下,骨髓源性內(nèi)皮祖細胞(EPCs)VEGF與SDF-1表達是否能增強。方法Percoll密度梯度離心法分離SD大鼠骨髓單個核細胞,血管內(nèi)皮生長因子(VEGF)、表皮生長因子(EGF)、堿性成纖維細胞生長因子(bFGF)誘導(dǎo)培養(yǎng),并進行形態(tài)學(xué)、免疫學(xué)、功能學(xué)(Dil-acLDL與FITC-UEA-1雙熒光染色)鑒定。將鑒定為EPCs的細胞分為單純EPCs組、EPCs+丹參酮IIA組,另
2、設(shè)單純丹參酮IIA組及對照組(PBS液)。細胞培養(yǎng)第9d,Westernblot與實時熒光定量PCR檢測各組VEGF、SDF-1基因與蛋白表達。結(jié)果EPCs+丹參酮IIA組VEGF、SDF-1基因與蛋白表達均高于其余各組(p均<0.05)。結(jié)論丹參酮IIA可上調(diào)大鼠骨髓源性內(nèi)皮祖細胞VEGF及SDF-1表達,從而可能在缺血環(huán)境中更好促進EPCs歸巢,更有效促進血管修復(fù)與再生。【關(guān)鍵詞】骨髓;內(nèi)皮祖細胞;丹參酮IIA;歸巢;血管新生【中圖分類號】R541.4【文獻標志碼】AEfficiencyofratbonemarrow-derivedendothelialp
