1、枸杞多糖對實驗性肝癌小鼠腫瘤細胞FasL表達的影響及其抗腫瘤作用機制作者:何彥麗,杜標炎,王慧鋒,蘇寧,肖霞【摘要】 【目的】觀察枸杞多糖(LBP)對實驗性肝癌小鼠腫瘤細胞死亡分子Fas配體(FasL)表達的影響,探討其抗腫瘤作用機制?!痉椒ā窟x用昆明種小鼠,隨機分為模型組,LBP低、高劑量組(劑量分別為0625、1250g·kg-1·d-1),各組均采用右側腋下接種H22肝癌腹水型細胞復制荷瘤模型,從第3天開始按設計劑量給藥,連續(xù)14d。采用蘇木素—伊紅(HE)染色觀察各組腫瘤細胞密度、細胞核分裂計數(shù)及間質淋巴細胞浸潤情況,采用免疫組化法觀察各組小鼠腫瘤細
2、胞FasL表達情況?!窘Y果】LBP低、高劑量組均可顯著降低腫瘤細胞核分裂計數(shù)(P<005);LBP高劑量組可升高腫瘤間質淋巴細胞浸潤程度(P<005),HE染色顯示在腫瘤邊緣組織內見多量淋巴細胞浸潤。LBP低、高劑量組均可顯著降低小鼠腫瘤細胞FasL陽性表達指數(shù)(P<005);免疫組化染色顯示模型組FasL表達強陽性,LBP低劑量組部分肝癌細胞內FasL表達陽性,FasL僅在LBP高劑量組少量肝癌細胞中有表達?!窘Y論】LBP抗實驗性肝癌的效應可能與其能抑制FasL表達,減少免疫活性細胞凋亡有關?!娟P鍵詞】枸杞多糖/藥理學肝腫瘤/中藥療法腫瘤組織/病理學基因表

4、H22bearingmicemodelswereinducedthroughrightsubaxillaryinoculationofH22asciticcells.Threedaysafterinoculation,LBPgroupweregivenLBPfor14days.Hematoxylinandeosin(HE)stainingmethodwasusedtoexaminethetumorcelldensity,tumorcellmitoticcount,andlymphocyteinfiltrationintumorinterstitialtissu
5、e.FasLexpressionwasobservedinthethreegroupswithimmunohistochemicalmethod.ResultsTumorcellmitoticcountwasdecreasedinthetwoLBPgroups(P<005),thescoreoflymphocyteinfiltrationintumorinterstitialtissuewasincreased,andlymphocyteinfiltrationwasobviousafterHEstaininginhighdoseLBPgroup(P&l
6、t;005).TheindexofFasLexpressionpositivewasdecreasedinbothLBPgroups(P<005).FasLexpressionwasstrongpositiveinthemodelgroup,andpositiveinpartiallivertumorcellsoflowdoseLBPgroupandinlesslivertumorcellsofhighdoseLBPgroup.ConclusionThe11antitumormechanismofLBPisprobablyrelatedwithth
