2、39基因突變菌株的生物學(xué)特性和毒力進(jìn)行研究.為進(jìn)一步明確毒力島基因PG0839的功能提供實(shí)驗(yàn)依據(jù)。方法:培養(yǎng)/9.gingivalisW83和PG0839基因突變菌株.觀察2個(gè)菌株菌落形態(tài)特征和革蘭染色鏡下特征,繪制菌株的生長曲線。制備1%綿羊紅細(xì)胞,取2種細(xì)菌細(xì)胞.進(jìn)行綿羊紅細(xì)胞凝集活性檢測(cè)。分別接種濃度為2xlOm、lxlOlO、5x10’CFU/ml的gin~valisW83和PG0839基因突變株菌液于小鼠背部采用SPSS13.0軟件包中的皮爾森卡方檢驗(yàn)和對(duì)數(shù)秩檢驗(yàn)對(duì)不同組別小鼠生存情況進(jìn)行分析。結(jié)果:與PgingivaliW83比較.PG0839突變菌株產(chǎn)生黑色素和紅
3、細(xì)胞凝集活性顯著降低。2個(gè)菌株生長曲線未見顯著差異(25.50,P=0.19)小鼠皮下感染模型表明,PG0839基因突變菌株毒性顯著低于W83菌株(P<0.05)。結(jié)論:PG0839基因的缺失改變了細(xì)菌的一些生物學(xué)特征,提示PG0839基因可能調(diào)控gingiva!is某些基因或者基因產(chǎn)物的表達(dá)同時(shí),PG0839基因在Pgingivalis致病過程中發(fā)揮一定作用。f關(guān)鍵詞】牙齦卟啉單胞菌;毒力島;紅細(xì)胞凝集活性;生存率『中圖分類號(hào)1R780.1『文獻(xiàn)標(biāo)志碼1AInitialstudyofbiologicalcharacterizationandvirulenceofPorphyr
