1、·540·ChinaTropicalMedicineMay2013Vol.13No.5中國熱帶醫(yī)學(xué)2013年第13卷第5期·論著·兩種方法分離小鼠脾原始T淋巴細胞效果比較*陳嶸祎,阿彩嶺,周英,樊翌明摘要:目的比較Ficoll-Paque梯度離心法與免疫磁珠法分離T淋巴細胞的效果差異,篩選有效的、優(yōu)異的分選小鼠脾原始T淋巴細胞方法。方法分別采用Ficoll-Paque梯度離心法及MiltenyiBiotec免疫磁珠進行小鼠淋巴細胞分離。血小板計數(shù)器統(tǒng)計單核淋巴細胞數(shù),苔盼藍染色判斷細胞存活率,流式細胞術(shù)鑒定CD4+CD62
2、+T淋巴細胞純度,并進行統(tǒng)計學(xué)分析。結(jié)果免疫磁珠法CD4+CD62+T淋巴細胞純度較高為(66.27±3.52)%,F(xiàn)icoll-Paque梯度離心法細胞純度為(15.27±4.16)%,兩者比較差異有統(tǒng)計學(xué)意義(P<0.01);但Ficoll-Paque梯度離心法獲取淋巴細胞數(shù)(1.47±0.35)×108較免疫磁珠法的(0.67±0.26)×108多,兩者比較差異有統(tǒng)計學(xué)意義(t=8.199,P<0.01),且Ficoll-Paque梯度離心法細胞存活率(93.9±2.6)%較免疫磁珠法的(89.9±2.9)%高,兩者
