2、logicetd'Immunologie,78850,Thiverval-Grignon,France(Accepted5June1981)SUMMARYThethermolabilityoftransmissiblegastroenteritisvirus(TGEV)wasstudiedbetween31and55°Cusingtwodifferentstrains.Thelossofinfectivityfollowedfirstorderkineticsexceptatthehighesttemperature.Thevaluesoft
3、hethermodynamicparametersindicatedthatthemechanismsinvolvedaboveandbelow45°Careclearlydistinct.TheratesofinactivationweregreateratalkalinethanatneutralpH,yetthenatureofthereactionappearedunchanged.Usingfourindependentstocksofmutagenizedvirus,wefailedtoselectthermal-resistan
4、tmutantsbysurvivorselectionat38°C.Incontrast,thermal-resistantmutantswereconsistentlyobtainedat54°C.However,thelatterdidnotshowanyincreasedstabilityat38°C,confirmingthefactthatadifferentinactivationprocesstakesplaceathighandatphysiologicaltemperatures.INTRODUCTIONEarlierstu
5、diesontheeffectofheatingRNAvirusesledtotheconclusionthatinactivationbetween30and60°Cgenerallytakesplaceaccordingtotwothermodynamicallydistinctmechanisms,dependingonthetemperaturerange.Infectivitydegradationat'low'temperatureisconsideredtobecausedselectivelybydenaturationoft
6、henucleicacid,whereasfunctionsrelatedtotheproteincomponentsoftheviruswouldbeprimarilylostat'high'temperature(Dimmock,1967;Ginoza,1968;Flemming,1971).Thepresentreportisconcernedwiththermalinactivationoftransmissiblegastroenteritisvirusofswine,amemberofthecoronaviridae.Thisfa
