1、中國獸醫(yī)學(xué)報2012年1O月第32卷第1O期Chin-,SciOct.2012Vo1.32No.101547苦馬豆素對新生SD大鼠大腦皮質(zhì)神經(jīng)細(xì)胞形態(tài)學(xué)損傷作用的研究龐龍,路浩,趙寶玉,張糅,王姍姍,宋巖巖,趙一楠(西北農(nóng)林科技大學(xué)動物醫(yī)學(xué)院,陜西楊凌712100)摘要:通過建立新生sD大鼠大腦皮質(zhì)神經(jīng)細(xì)胞原代體外培養(yǎng)模型,采用MTT法測定一個生長周期內(nèi)神經(jīng)細(xì)胞的活性分布,運用寇氏法計算苦馬豆素對大鼠大腦皮質(zhì)神經(jīng)細(xì)胞的Ic。。然后用不同質(zhì)量濃度苦馬豆素染毒12h,倒置相差顯微鏡觀察神經(jīng)細(xì)胞形態(tài)并測量其胞體短直徑和突起長度的變化。結(jié)果顯示,一個生長周期內(nèi)的大鼠大腦皮
2、質(zhì)神經(jīng)細(xì)胞活力最強(qiáng)為培養(yǎng)第5天,苦馬豆素對大鼠大腦皮質(zhì)神經(jīng)細(xì)胞的Ic。為0.8511g/L。與對照組相比,各試驗組神經(jīng)細(xì)胞密度降低,神經(jīng)網(wǎng)絡(luò)減少;除0.5g/L試驗組外,其他各試驗組神經(jīng)細(xì)胞胞體短直徑顯著減小(P0.05)。結(jié)果表明,苦馬豆素能明顯影響新生大鼠大腦皮質(zhì)神經(jīng)細(xì)胞的生長發(fā)育,苦馬豆素作用質(zhì)量濃度與神經(jīng)細(xì)胞形態(tài)改變存在明顯劑量一效應(yīng)關(guān)系。關(guān)鍵詞:苦馬豆素;大腦皮質(zhì)神經(jīng)細(xì)胞;形態(tài)學(xué);新生大鼠中圖分類號:$852.16文獻(xiàn)標(biāo)志碼:A文章編號:1005—4545(2012)10—1547—0
