1、JSouthMedUniv,2017,37(10):1405-1409doi10.3969/j.issn.1673-4254.2017.10.21·1405·基礎(chǔ)研究???????????????????????廖芷吟,趙柯湘,肖謙重慶醫(yī)科大學(xué)附屬第一醫(yī)院老年病科,重慶400016摘要:目的觀察白藜蘆醇對老年大鼠骨骼肌質(zhì)量、抓力、肌原纖維超微結(jié)構(gòu)以及AMPK/sirt1通路的影響。方法20只25月齡SD大鼠隨機(jī)分為老年對照組(OC組,10只)和老年白藜蘆醇組(OR組,10只),另外10只6月齡SD大鼠作為青年對照組(YC組)。OR組給予連續(xù)6周白藜蘆醇干預(yù)。電子抓力儀測量大鼠前肢抓力,
2、分析天平稱量大鼠腓腸肌質(zhì)量,用腓腸肌質(zhì)量(mg)/體質(zhì)量(g)得出肌肉減少癥指數(shù)(SI),透射電子顯微鏡觀察大鼠肌原纖維肌小節(jié)、明帶、暗帶以及H帶長度的改變。Westernblot方法檢測AMPK、P-AMPK、sirt1的蛋白表達(dá)量。結(jié)果OC組腓腸肌SI值低于YC組(P<0.05),抓力低于YC組(P<0.05),肌小節(jié)長度、暗帶長度、明帶長度和H帶長度均高于YC組(P<0.05),AMPK、P-AMPK、sirt1的蛋白表達(dá)量均低于YC組(P<0.05);與OC組相比,OR組SI值未見顯著性差異(P>0.05),抓力升高(P<0.05),肌小節(jié)長度、明帶長度和H帶長度均降低(P<0
3、.05),暗帶長度未見統(tǒng)計(jì)學(xué)差異(P>0.05),P-AMPK、sirt1的蛋白表達(dá)量均顯著增高(P<0.05)。結(jié)論白藜蘆醇不能減緩肌肉質(zhì)量的減少,但是可以增加老年大鼠的抓力。這種作用可能跟白藜蘆醇激活A(yù)MPK/sirt1通路,改善肌原纖維結(jié)構(gòu)的老化有關(guān)。關(guān)鍵詞:白藜蘆醇;肌肉減少癥;抓力;電鏡EffectofresveratrolonforelimbgripstrengthandmyofibrilstructureinagedratsLIAOZhiyin,ZHAOKexiang,XIAOQianDepartmentofGeriatrics,FirstAffiliatedHospit
