2、建立大鼠肝纖維化模型。在造模同時(shí),各給藥組每天給予相應(yīng)藥物10mL/(kg·d11次,連續(xù)給藥l1周。于11周末處死大鼠,免疫組化法檢測(cè)肝組織中Ot一平滑肌肌動(dòng)蛋白(a.SMA)的表達(dá),反轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)法檢測(cè)大鼠肝組織中轉(zhuǎn)化生長(zhǎng)因子(TGF.13)及smad3基因的表達(dá)。結(jié)果與對(duì)照組比較,模型組大鼠肝組織0【.SMA蛋白及TGF.D1、smad3基因的表達(dá)顯著上調(diào)(P<0.05)。與模型組比較,鱉甲煎丸和秋水仙堿可顯著降低大鼠肝組織c【一SMA蛋白及TGF.B6、smad3基因的表達(dá),其中以鱉甲煎丸2.2g/kg的效果更顯著。結(jié)論鱉甲煎丸抗肝纖維化作
