1、卷柏降糖作用的實驗研究摘要:目的:研究了不同提取方法對卷柏降糖活性的影響,并進一步探討了卷柏降糖作用的有效部位。方法:首先,采用HepG2細胞體外細胞篩選方法,觀察卷柏醇提物和卷柏水提物兩種不同提取方法對HepG2細胞培養(yǎng)液中葡萄糖消耗量的影響,并用四甲基偶氮唑鹽(MTT)方法檢測HepG2細胞的增殖;同時結(jié)合整體動物實驗的方法,比較了醇提物和水提物對高糖高脂飼料加小劑量鏈脲佐菌素(STZ)造成的大鼠糖尿病模型的降糖活性,以及對糖尿病大鼠葡萄糖耐量的影響。進而將卷柏醇提物上聚酰胺柱所得不同部位流
2、份,以HepG2細胞葡萄糖消耗實驗作為指標對降糖的有效部位進行了探討,并采用MTT法檢測受試藥物對HepG2細胞增殖的影響。結(jié)果:體外實驗結(jié)果顯示:醇提物和水提物在一定的濃度范圍內(nèi)均能顯著促進HepG2細胞的葡萄糖消耗,醇提物比水提物作用活性強;整體動物實驗結(jié)果顯示:卷柏醇提物和水提物均能降低糖尿病大鼠的血糖,并能在一定程度升高糖尿病大鼠的葡萄糖耐量,且卷柏醇提物降糖作用明顯優(yōu)于卷柏水提物;卷柏活性部位的的篩選結(jié)果顯示:卷柏50%EtOH部位能夠顯著促進培養(yǎng)液中HepG2細胞的葡萄糖消耗,與正常
3、組比較有極顯著性差異(P<0.01),水部位和80%EtOH部位次之,95%EtOH部位作用不明顯。結(jié)論:細胞實驗結(jié)合動物實驗結(jié)果提示醇提物比水提物具有更好的降糖效果;卷柏醇提物上聚酰胺柱所得50%EtOH部位體外降糖活性最高。關(guān)鍵詞:卷柏;HepG2細胞;糖尿病大鼠;葡萄糖消耗;血糖AntihyperglycemicactivityofSelaginellaTamariscinaABSTRACT:OBJECTIVE:ToinvestigatethehypoglycemiceffectsofSe

7、creasetheglucoseconsumptiononHepG2cellswithincertainconcentration,andtheeffectsofEtOHextractwerehigherthanH2Oextract;TheresultsinvivoindicatethatboththeEtOHandH2Oextractscanlowerthefastingbloodglucoselevelsandimproveoralglucosetoleranceondiabeticrats
