6、.2.5EffectofantifungalsubstanceofT111onmembraneosmoticpotential184Discussion195Reference206Thanks21放線菌T111發(fā)酵液理化性質(zhì)及發(fā)酵產(chǎn)物對(duì)核盤菌抑菌機(jī)制初探作者:韓紅寧(06植檢一)指導(dǎo)教師:慕衛(wèi)摘要:為明確放線菌T111的理化性質(zhì),研究了其抗菌譜,及貯存時(shí)間、溫度、酸堿條件、紫外光照射等對(duì)其發(fā)酵液穩(wěn)定性的影響。結(jié)果表明,T111發(fā)酵液對(duì)黃瓜菌核、黃瓜蔓枯、棉花紅腐病菌等有較高的抑制活性;貯存穩(wěn)定性和熱穩(wěn)定性良好,4℃貯存60d抑菌活性為對(duì)照的88.3%,80℃處理60min抑菌活性為對(duì)
7、照的90.9%;對(duì)pH值較敏感,在pH5~7時(shí)活性較高;對(duì)紫外線穩(wěn)定,紫外光照射12h,其活性基本不受影響。初步研究了T111發(fā)酵產(chǎn)物對(duì)核盤菌的抑菌機(jī)制,采用菌絲生長(zhǎng)速率法測(cè)定了其對(duì)黃瓜菌核病菌Sclerotiniasclerotiorum菌絲生長(zhǎng)的毒力,及其對(duì)菌絲形態(tài)、菌核形成、菌核萌發(fā)和菌體細(xì)胞膜透性的影響。結(jié)果表明,T111發(fā)酵產(chǎn)物對(duì)核盤菌菌絲生長(zhǎng)的EC50值為12.5μg/mL,低于對(duì)照藥劑乙霉威及嘧霉胺;經(jīng)發(fā)酵產(chǎn)物處理后,菌絲形態(tài)出現(xiàn)
