5、································26中文摘要青皮中橙皮苷提取及純度測定陳秀秀河北北方學(xué)院醫(yī)學(xué)院藥學(xué)系藥學(xué)專業(yè)07級目的:通過優(yōu)化文獻(xiàn)提取橙皮苷的方法得到提取橙皮苷的最佳工藝條件并采用TCL法進(jìn)行定性鑒別(2010版藥典第一部);采用高效液相色譜法(HPLC)測定橙皮苷的純度。方法:以青皮為原料,用堿液和甲醇做溶劑提取橙皮苷的條件為:粗苷:甲醇:堿液[NaOH濃度為0.1%]=1:23:47在85oC以上浸泡4h,調(diào)節(jié)pH值為4。即先將甲醇和堿液攪拌混合,再將粗苷加入,繼續(xù)攪拌1h使粗苷溶解完全,然后過濾

6、至濾液澄清,用鹽酸調(diào)濾液的pH值至4,靜置沉淀,過濾,濾餅用煮沸過的水洗滌至無色,甲醇濾液精餾回收,橙皮苷的收率達(dá)85%以上。TCL鑒別出與橙皮苷對照品相對應(yīng)的寬點(diǎn);HPLC法測定出橙皮苷純度為85.94%且理論塔板數(shù)不低于1000。結(jié)論:此方法操作簡便,易控制且提取橙皮苷的效率較高,所建立的定性定量方法專屬性強(qiáng),重復(fù)性好。關(guān)鍵詞:青皮;橙皮苷;薄層色譜法;紫外分光光度法;高效液相色譜法AbstractExtractionandpurificationofhesperidinincicusprocessingandmeasuringi
