1、中國醫(yī)藥導(dǎo)報(bào)2014年3月第1l卷第7期論著·丹皮酚對結(jié)腸癌細(xì)胞系LoVo細(xì)胞增殖、凋亡的影響及其機(jī)制李明譚詩云武漢大學(xué)人民醫(yī)院消化內(nèi)科.湖北武漢430060【摘要】目的觀察丹皮酚對人結(jié)腸癌細(xì)胞系LoVo細(xì)胞增殖、凋亡的影響及探討可能的作用機(jī)制。方法用不同濃度丹皮酚處理體外培養(yǎng)的IJoV0細(xì)胞24、48、72、96h,CCK一8比色法測定其對結(jié)腸癌LoVo細(xì)胞的活力的影響:倒置顯微鏡下觀察細(xì)胞的形態(tài)學(xué)變化;根據(jù)CCK一8結(jié)果分為丹皮酚組(濃度31.25、62.50、125mg/L)和對照組,處理48h后,流式細(xì)胞儀檢測細(xì)胞凋亡;激光共聚焦顯微鏡檢
2、測細(xì)胞內(nèi)Ca濃度的變化;RT—PCR和Westernblot法檢測RUNX3基因表達(dá)情況。結(jié)果丹皮酚能顯著降低LoVo細(xì)胞存活率,呈劑量、時(shí)問依賴性;倒置顯微鏡下可觀察到典型的細(xì)胞凋亡形態(tài);丹皮酚組(濃度31.25、62.50、125mg/L)處理細(xì)胞48h后.凋亡率分別為(12-3±1.3)%、(22.4+1.5)%、(36.2+2.1)%,與對照組[凋亡率(2.3±0.9)%]比較,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05);丹皮酚組處理48h后,細(xì)胞鈣離子熒光強(qiáng)度分別為(41.36_+4.62),(57.51+3.83)和(69.43~3.76),
