2、中選育出的材料24-6進(jìn)行了形態(tài)學(xué)觀察、細(xì)胞學(xué)檢測(cè)和原位雜交(GISH)分析。結(jié)果表明:(1)通過(guò)根尖細(xì)胞學(xué)鑒定,該材料染色體數(shù)為42條,2n=42;(2)以華山新麥草Ns基因組DNA為探針,普通小麥7182基因組DNA為封阻,根尖與花粉母細(xì)胞原位雜交(GISH)顯示該材料攜帶有2條可以配對(duì)的華山新麥草染色體臂,確定24-6為穩(wěn)定的小麥-華山新麥草易位系;(3)田間觀察發(fā)現(xiàn)24-6植株緊湊,葉色深綠,籽粒白色、呈半角質(zhì),比其母本(普通小麥7182)株高略有降低,穗長(zhǎng)、小穗數(shù)、穗粒數(shù)、千粒重、分蘗數(shù)都有所增加。關(guān)鍵詞:華山新麥草;遠(yuǎn)緣雜交;易位系;基因組原位雜交(GISH)中圖分類號(hào):S5
