1、姜黃素對慢性應(yīng)激大鼠腎上腺、血清ACTH和免疫功能的影響【摘要】目的:探討姜黃素對慢性不可預(yù)知應(yīng)激模型大鼠的腎上腺結(jié)構(gòu)、血清促腎上腺皮質(zhì)激素(ACTH)濃度和免疫功能的影響及其意義。方法:采用多種不可預(yù)知的刺激方式交替、持續(xù)應(yīng)激20d建立大鼠慢性應(yīng)激模型。隨機(jī)分為正常對照組、應(yīng)激模型組、姜黃素組(2.5mg/kg、5mg/kg、10mg/kg)、丙咪嗪組(10mg/kg),每組6只。采用HE染色技術(shù),觀察應(yīng)激大鼠腎上腺結(jié)構(gòu)的改變及姜黃素的作用。通過測定應(yīng)激大鼠外周血中白細(xì)胞(WBC)數(shù)量和血清中ACTH和白細(xì)胞介素1β(Int
2、erleukin-1β,IL-1β)含量的改變,觀察姜黃素的作用。結(jié)果:慢性應(yīng)激大鼠與正常對照組比較腎上腺皮質(zhì)增厚,髓質(zhì)萎縮,外周血白細(xì)胞數(shù)量明顯減少(包括淋巴細(xì)胞、單核細(xì)胞和中性粒細(xì)胞百分比下降),血清中ACTH和IL-1β含量明顯增加。姜黃素組大鼠腎上腺結(jié)構(gòu)變化不明顯,外周血WBC數(shù)量不同程度增加,血清中ACTH和IL-1β含量降低。結(jié)論:姜黃素可能通過調(diào)節(jié)下丘腦-垂體-腎上腺軸以及免疫系統(tǒng)功能,在慢性應(yīng)激大鼠模型中顯示出抗抑郁活性。【關(guān)鍵詞】姜黃素應(yīng)激抗抑郁腎上腺白細(xì)胞計數(shù)ACTHIL-1βAbstract:Object

5、alsodetectedusingtheenzymelinkedimmunosorbentassay.Results:Chronicstressresultedindiffusehyperplasiaoftheadrenalcortexandatrophyoftheadrenalmedullainrats,andtheserumlevelofACTHwasincreased,whichsuggestedthatstress-adaptionfailureoftheadrenalglandoccurred,whileadrena
6、lglandoftheratspretreatedwithcurcumin(10mg/kg)priortoeachstressorfor21daysdidnotresultinanypathologicchangesandthelevelofACTHdecreased.Moreover,chronicstressalsosignificantlyreducedtheWBCcountandrelativeWBCpercentagesintheperipheralblood,includingthepercentageoflymp
7、hocytes,monocytes,neutrophils,however,curcuminreversedthesechangesandraisedWBCcount,alongwithrelativeWBCpercentagessignificantly.Meanwhile,theserumlevelofIL-1βwasincreasedinchronicallystressedanimals,andcurcuminalso9declinedthelevelofthisstresshormone.Conclusion:The
8、seresultssuggestthatcurcuminexertsantidepressanteffectsagainstchronicstress,andthemechanismmaybemediatedbyregulatingthefunctionofthehypoth
