1、川芎嗪對高糖環(huán)境大鼠腎小球系膜細胞增殖及其細胞外基質(zhì)含量的影響【摘要】目的:探討川芎嗪對高糖環(huán)境下大鼠腎小球系膜細胞(MCs)增殖及其細胞外基質(zhì)含量的影響。方法:體外培養(yǎng)大鼠腎小球系膜細胞株,分對照組、高糖組、TMP組,分別培養(yǎng)12h、24h、48h,CASY細胞計數(shù)儀計數(shù)細胞存活率;以CCK-8法檢測細胞增殖;以ELISA法檢測細胞上清液細胞外基質(zhì)ColIV及FN的含量。結(jié)果:各TMP濃度組之間存活率無統(tǒng)計學差異(P>0.05);與對照組比較,高糖下培養(yǎng)24h、48hMCs增殖顯著增快(P<0.01),分泌的ColIV及FN含量增多(P<0.05);與高糖組比

7、環(huán)、血液流變學,降血糖、抗氧化、抗纖維化和拮抗鈣離子等藥理作用,已廣泛應用于心、腦、肺、腎及血管疾病的治療,大量臨床及動物實驗證明TMP對DN有很好的療效。本研究觀察了TMP對高糖環(huán)境下大鼠腎小球系膜細胞(ratmesangialcells,MCs)增殖及FN、ColIV含量的影響,進一步探討TMP對糖尿病腎病的保護作用及其可能機制。1材料和方法1.1材料大鼠腎小球系膜細胞株購自上海長征醫(yī)院;鹽酸川芎嗪粉劑購自山東長富潔晶藥業(yè)有限公司;正常葡萄糖(含5.5mmol/LD-葡萄糖)
