1、補腎柔肝方對二甲基亞硝胺誘導大鼠肝纖維化的預防作用及其機制研究【摘要】探討補腎柔肝方對二甲基亞硝胺(dimethylnitrosamine,DMN)誘導大鼠肝纖維化的預防作用及初步機制。方法:雄性Wistar大鼠40只,隨機分為正常對照組(n=10)、模型組(n=15)及補腎柔肝方預防組(n=15)。模型組和補腎柔肝方預防組以10mg/kgDMN腹腔注射,1次/d,每周連續(xù)3d,共4周。模型組在造模同時給予生理鹽水灌胃,補腎柔肝方預防組在造模同時給予補腎柔肝方灌胃,共4周;第4周末處死全部大鼠,留取血清及肝組織標本。采用全
2、自動生化分析儀檢測血清總膽紅素(totalbilirubin,Tbil)、丙氨酸氨基轉移酶(alanineaminotransferase,ALT)、天冬氨酸氨基轉移酶(aspartateaminotransferase,AST)、白蛋白等;應用逆轉錄聚合酶鏈式反應法檢測肝組織結締組織生長因子(connectivetissuegrowthfactor,CTGF)和Ⅰ型膠原mRNA的表達。結果:補腎柔肝方預防組大鼠一般狀態(tài)明顯好于模型組;模型組大鼠死亡率為13.33%(2/15),補腎柔肝方預防組死亡率為6.67%(1/15
3、);模型組大鼠腹水發(fā)生率為46.67%(7/15),補腎柔肝方預防組大鼠腹水發(fā)生率為20%(3/15),兩組比較差異有統(tǒng)計學意義(P<0.01);補腎柔肝方組肝功能較模型組有顯著改善(P<0.01)。模型組肝組織CTGF和Ⅰ型膠原mRNA表達明顯增強,補腎柔肝方預防組與模型組相比明顯減弱(P<0.01)。結論:補腎柔肝方可通過抑制大鼠肝組織CTGF和Ⅰ型膠原mRNA的表達,達到抗肝纖維化的作用?!娟P鍵詞】補腎柔肝方肝纖維化二甲基亞硝胺結締組織生長因子Ⅰ型膠原大鼠Objective:Tostudytheef

7、reversetranscriptionpolymerasechainreaction.6Results:ItwasfoundthattheserumTbillevelandtheactivitiesofAST,ALTweredeclinedintheBSRGRgroupascomparedwiththoseintheuntreatedgroup(P<0.01).TheserumalbumincontentintheBSRGRgroupwasincreasedascomparedwiththatintheuntrea
8、tedgroup(P<0.01).TheexpressionsofcollagentypeⅠandCTGFmRNAsintheuntreatedgroupwerehigherthanthoseintheBSRGRgroup(P<0.01).Conclusion:BSRGRcandecreas
