6、······························50個(gè)人簡歷·······································································································51新型復(fù)合支架對(duì)骨髓間充質(zhì)干細(xì)胞增殖及分化影響的研究摘要目的:創(chuàng)傷、退行性病變或腫瘤等原因?qū)е碌墓侨睋p仍然是骨骼修復(fù)重建的主要難題。同種異體骨移植和自體骨移植是當(dāng)前治療骨缺損的最佳手段。而這兩種治療都有其局限性,如供體組織有限、供體部位發(fā)病率高、手術(shù)復(fù)雜、同種異體移植物疾

7、病傳播或免疫排斥的可能性、且移植物無法準(zhǔn)確塑形替代原組織的生物力學(xué)特性。如何實(shí)現(xiàn)骨缺損修復(fù)重建一直是當(dāng)今醫(yī)學(xué)界的難題,組織工程支架為解決這一難題提供了很好的思路。本實(shí)驗(yàn)旨在采用冷凍干燥法制備膠原蛋白/殼聚糖/羥基磷灰石三維仿生骨組織工程支架,并觀察膠原蛋白/殼聚糖/羥基磷灰石三維復(fù)合支架對(duì)骨髓間充質(zhì)干細(xì)胞的增殖和分化的影響。目的是研究組織工程支架治療骨缺損修復(fù)重建的可行性。方法:1.大鼠骨髓間充質(zhì)干細(xì)胞(BMSCs)的分離培養(yǎng)和分化鑒定:抽取清潔級(jí)健康雄性SD大鼠骨髓,密度梯度離心法提取單個(gè)核細(xì)胞,貼壁法分離骨髓間充質(zhì)干細(xì)胞。通過CCK-8法檢測(cè)

8、細(xì)胞生長曲線;成骨誘導(dǎo)分化培養(yǎng)后,進(jìn)行茜素紅染色檢測(cè)礦物結(jié)節(jié)染色評(píng)估成骨分化情況。2.膠原/殼聚糖/羥基磷灰石復(fù)合支架制備:采用冷凍干燥
