2、expressioncaninhibittheinvasionabilityofMHCC97hepatocellularcarcinomacells.【Keyacell;invasion肝癌嚴(yán)重威脅著人類的健康,在中國肝癌的發(fā)病率一直呈逐年上升趨勢。我國每年約11萬人死于肝癌.freela公司。Matrigel膠購于BDBiosciences公司。1.2OligofecTAMINE介導(dǎo)的反義FAKODN轉(zhuǎn)染反義FAKODN5’-ATAATCCAGCTTGAACCAAG-3’。由上海生工生物工程公司DNA合成,全硫代
3、修飾,聚丙烯酰胺凝膠電泳純化,凍存。OligofecTAMINE按產(chǎn)品說明書介導(dǎo)轉(zhuǎn)染培養(yǎng)細(xì)胞。對照組只加入脂質(zhì)體不加入反義FAKODN。1.3Genistein抑制FAK磷酸化以10μg/ml的終濃度加入細(xì)胞培養(yǎng)液中,并使DMSO終濃度為0.4%。實(shí)驗(yàn)對照組以同樣終濃度為0.4%的溶劑處理。1.4細(xì)胞體外侵襲能力檢測法Trans孔徑)黏附于Transl,加入濾膜上表面(5μg/濾膜),室溫下通風(fēng)櫥中干燥過夜。用前Matrigel以40μl無血清培養(yǎng)基重建20min后備用。將行不同預(yù)處理后的細(xì)胞用無血清培養(yǎng)基洗3次,
4、0.25%胰蛋白酶消化,重懸于含0.1%BSA的培養(yǎng)基中,取2×106/ml細(xì)胞100μl加入至上室,600μl0.1%小牛血清培養(yǎng)基加入下室。37℃、5%CO2條件下培養(yǎng)20h后,濾膜用甲醇固定5min,行Gimsa染色。去除濾膜上層細(xì)胞,顯微鏡下(×320)計(jì)數(shù)濾膜下表面細(xì)胞數(shù),隨機(jī)計(jì)數(shù)5個視野中的細(xì)胞數(shù)目,每個標(biāo)本重復(fù)2次,實(shí)驗(yàn)重復(fù)2次,以侵襲細(xì)胞的相對數(shù)目來表示腫瘤細(xì)胞的侵襲能力。1.5統(tǒng)計(jì)學(xué)方法采用t檢驗(yàn)。2結(jié)果我們發(fā)現(xiàn)轉(zhuǎn)染反義FAKODN和經(jīng)Genistein處理后的MHCC97肝癌細(xì)胞穿透Matrig
