1、血晶素對(duì)缺氧缺糖海馬腦片的保護(hù)作用及其分子機(jī)制王莉1,余剛2,向德兵3,朱潔2,李光勤21.重慶市腫瘤研究所,重慶400030;2.重慶醫(yī)科大學(xué)附屬第一醫(yī)院神經(jīng)內(nèi)科,重慶4000163:第三軍醫(yī)大學(xué)大坪醫(yī)院野戰(zhàn)外科研究所病理科,重慶400042摘 要: 目的觀察血晶素(Hemin)對(duì)海馬腦片氧糖剝離(OGD)模型的保護(hù)作用并探討其機(jī)制。方法:取新生8-10dSD大鼠,制備厚約400μm海馬腦片,培養(yǎng)2周后篩取完好無特異熒光著色的腦片,分為正常培養(yǎng)對(duì)照組(HOTC),缺氧缺糖組(OGD),預(yù)加Hemin再缺氧缺
2、糖組(Hemin+OGD),預(yù)加血紅素加氧酶-1(hemeoxygenase-1,HO-1)抑制劑鋅原卟啉(Znpp)再缺氧缺糖組(Znpp+OGD)。除HOTC組外,其余各組在5%CO2、95%N2的混合氣體中培養(yǎng)1.5h,其后恢復(fù)培養(yǎng)24h。海馬腦片冰凍連續(xù)冠狀切片,應(yīng)用免疫組織化學(xué)染色和圖像分析處理技術(shù)觀察海馬腦片腦紅蛋白(neuroglobin,Ngb)和HO-1表達(dá)變化,并觀察神經(jīng)元形態(tài)學(xué)改變。結(jié)果:海馬腦片發(fā)生缺氧缺糖時(shí),神經(jīng)元中Ngb蛋白和HO-1蛋白表達(dá)增加,血晶素促進(jìn)神經(jīng)元Ngb蛋白和HO-
