1、摘要胚胎干細(xì)胞(EScell)是具有多向分化能力的全能性細(xì)胞。研究和利用ES細(xì)胞是當(dāng)前生物工程領(lǐng)域的核心問(wèn)題之一。至今建立的小鼠ES細(xì)胞系大多來(lái)源于129,C57BL/6J,BALA/C等品系,有關(guān)昆明白小鼠ES細(xì)胞建系研究較少。作為國(guó)內(nèi)普遍使用的一種實(shí)驗(yàn)動(dòng)物,探討昆明白小鼠胚胎干細(xì)胞建系方法更具應(yīng)用廣泛性。本實(shí)驗(yàn)研究了內(nèi)細(xì)胞團(tuán)(IcM)獲取方法,培養(yǎng)系統(tǒng),傳代方法等胚胎干細(xì)胞建系過(guò)程中必不可少的技術(shù)環(huán)節(jié),探索了大鼠心肌細(xì)胞條件培養(yǎng)基培養(yǎng)胚胎干細(xì)胞效果;對(duì)比了不同飼養(yǎng)層對(duì)胚胎干細(xì)胞集落形成的影響及維持能力,結(jié)果表明:自制的兔抗小鼠多克隆抗體能有效去除小鼠囊胚的滋養(yǎng)層;將獲得
2、的ICM在小鼠胚胎成纖維細(xì)胞(PMEF)飼養(yǎng)層且添加白血病抑制因子(LIF)條件下培養(yǎng),胚胎干細(xì)胞傳代效果最佳,體外培養(yǎng)已傳至第8代;所培養(yǎng)的ES細(xì)胞具有巢狀集落形態(tài),和周圍飼養(yǎng)層細(xì)胞存在明顯界線,集落內(nèi)部細(xì)胞間連接緊密;ES集落經(jīng)堿性磷酸酶染色呈強(qiáng)陽(yáng)性;體外分化檢測(cè)發(fā)現(xiàn)培養(yǎng)的胚胎干細(xì)胞仍具有較強(qiáng)的多能性:對(duì)其進(jìn)行冷凍及復(fù)蘇實(shí)驗(yàn),解凍后ES細(xì)胞可以存活并生長(zhǎng)增殖。此外,本論文還就干細(xì)胞研究的發(fā)展過(guò)程、技術(shù)進(jìn)步、存在的問(wèn)題以及研究趨勢(shì)等方面作了概括性的綜述。關(guān)鍵詞:昆明白小鼠,胚胎干細(xì)胞,內(nèi)細(xì)胞團(tuán),、堿性磷酸酶染色THEESCELLLINEESTABLISⅢNGANDCHAR
