2、on;isolationofthetargetpigment;identificationofthepigment;stabilitytestsofthepigment.HPLC-MSanalysisshowedthatthenaturalpigmentinmyceliaofRCEF4585wasamixture.WiththehelpofhighresolutionMS,themolecularformulaofthemainpigmentwasidentifiedasC26H32O9.UVanalysisresultsrevealedthatthiscompoundhasultra-v
3、ioletabsorbanceat228nm、260nm、413nm、434nm.ThemainpigmentwaspossiblyanewcompoundaccordingthesearchingresultsofChapman&Hallnaturalproductsdatabase.TherewasonlyonepigmentfoundfromthemethanolextractsofthemyceliaofRCEF3669.AnalysisresultsofHPLC-DAD-MSrevealedthatthemolecularformulaofthepigmentwasC30H18O
4、12,andthattheultra-violetabsorbanceofthepigmentwas225nm、266nm、297nm、467nm.Thepigmentcompoundwasidentifiedasdimericanthraquinone,accordingtotheMsandUVdata.Thispigmentwasthefirsttimeisolatedfromentomogenousfungi.HPLC-MSanalysisshowedthatthenaturalpigmentsinthemethanolextractsfrommyceliaofRCEF4337wer
5、esimilartothosepigmentsfromRCEF4584.However,onedifferentpigmentwithmolecularformulaC30H18O13wasfoundinethylacetateextractsfromRCEF4337.Thepigmenthasultra-violetabsorbanceat225nm、266nm、297nm、503nm.Accordingtomolecularformulaandultra-violetabsorbance,thesearchingresultsofChapman&Hallnaturalproductsd
6、atabaserevealedthatthispigmentwaspossiblyanewcompound.High-speedcounter-currentchromatographywasusedforseparationandpurificationofnaturalpigmentP-1fromthemyceliaofRCEF4585.Thetow-phasesolventsystemwasethylacetate-n-butanol-methanol-water(10:3:3:10).Theupperphasewasstationaryphaseandthelowerphasewa
7、smobilephase.Theflow-rateofthemobilephasewas2.0ml/minandtherotationspeedwas850r/min.TheeffluentofthecolumnwasmonitoredwithaUVdetectorat340nm.AndSemi-PreparationHPLCwasemployedtoseparateandpurifynaturalpigment,tow
