3、結(jié)腸(編輯:宋威)炎發(fā)病的關(guān)系[J].中華醫(yī)學(xué)雜志,2011,91(23):1605-1608.左歸丸對(duì)衰老大鼠骨髓間充質(zhì)干細(xì)胞成骨和成脂分化的影響黃進(jìn),徐志偉,詹菲,張晨,丁富平,羅川晉,李杰斌,張進(jìn)(廣州中醫(yī)藥大學(xué),廣東廣州510405)摘要:目的研究左歸丸對(duì)衰老大鼠骨髓間充質(zhì)干細(xì)胞(MSCs)成骨與成脂分化的影響。方法通過(guò)腹腔注射D-半乳糖建模,制備衰老大鼠模型,分離培養(yǎng)MSCs,并誘導(dǎo)MSCs成骨和成脂分化。通過(guò)堿性磷酸酶(ALP)活性檢測(cè)、ALP染色觀察MSCs成骨分化能力,通過(guò)甘油三酯(TG)表達(dá)量和油紅O染色觀察MSCs成脂分化能力。結(jié)果模型組MSCsALP染色陽(yáng)性率明顯
4、低于正常組,左歸丸組MSCs染色陽(yáng)性率明顯高于模型組(P<0.05)。成骨誘導(dǎo)14d,模型組MSCsALP活性值明顯低于正常組而左歸丸組明顯高于模型組(P<0.05)。油紅O染色,模型組MSCs染色陽(yáng)性率明顯高于正常組,左歸丸組MSCs染色陽(yáng)性率明顯低于模型組(P<0.05)。成脂誘導(dǎo)7d,模型組MSCsTG表達(dá)量明顯高于正常組而左歸丸組MSCsTG表達(dá)量明顯低于模型組(P<0.05)。結(jié)論左歸丸可促進(jìn)衰老大鼠MSCs向成骨細(xì)胞分化,同時(shí)可抑制其向脂肪細(xì)胞分化。關(guān)鍵詞:左歸丸;衰老;骨髓間充質(zhì)干細(xì)胞;骨向分化;脂向分化中圖分類號(hào):R285.5文獻(xiàn)標(biāo)志碼:A文章編號(hào):1003-9783(
