1、摘要獨(dú)角蓮作為一種名貴中草藥,具有較高的醫(yī)療價(jià)值和防病保健功能,將其塊莖提取液應(yīng)用于抗腫瘤研究,并探討其相關(guān)機(jī)制。本文以東北產(chǎn)獨(dú)角蓮塊莖為原料,以蒸餾水做提取溶劑.以溶劑量是藥材的10倍為原則進(jìn)行加量提取,提取方式采用超聲波,時(shí)間為20min,間隔lmin后再重復(fù)一次。對(duì)真空干燥后得到獨(dú)角蓮塊莖提取物的活性分析表明,對(duì)酪氨酸酶的相對(duì)抑制率51.21%,達(dá)到最佳效果。MTT實(shí)驗(yàn)結(jié)果表明,獨(dú)角蓮塊莖提取液對(duì)體外Hela的增殖有一定的抑制作用:對(duì)Hcla的抑制率為65.08%。細(xì)胞凋亡實(shí)驗(yàn)和細(xì)胞周期流
2、式細(xì)胞儀檢測(cè)表明,獨(dú)角蓮塊莖提取液能引起凋亡細(xì)胞的增加,并成劑量依賴性,獨(dú)角蓮塊莖提取液作用72h后的凋亡率最高,為24.5%。Westernblot檢測(cè)HeLa細(xì)胞經(jīng)濃度遞增的塊莖提取液處理48h后,caspasc3蛋白表達(dá)逐漸增強(qiáng)。獨(dú)角蓮塊莖提取液對(duì)癌細(xì)胞的生長(zhǎng)抑制呈現(xiàn)出良好的量效關(guān)系。對(duì)接種$180細(xì)胞株形成實(shí)體瘤的小鼠用獨(dú)角蓮塊莖提取液灌胃表明,獨(dú)角蓮有顯著的抗腫瘤活性,抑瘤率為31.77%。對(duì)腹腔接種EAC細(xì)胞株形成腹水癌的昆明種小鼠用獨(dú)角蓮提取液灌胃表明獨(dú)角蓮能延長(zhǎng)腹水癌小鼠生存期,
