1、BMSCs分離培養(yǎng)、純化及鑒定作者:馬民呂志剛杜以寬張桂娟馬義【摘要】目的建立一種持續(xù)、穩(wěn)定的可多向分化的骨髓間充質(zhì)干細(xì)胞(BMSCs)體外分離培養(yǎng)體系。方法運(yùn)用密度梯度離心法從1月齡新西蘭兔骨髓中分離培養(yǎng)BMSCs,利用相差顯微鏡觀察其形態(tài)及生長(zhǎng)情況,掃描電鏡觀察其細(xì)胞結(jié)構(gòu),運(yùn)用流式細(xì)胞術(shù)分析分離細(xì)胞群所處細(xì)胞周期和細(xì)胞活力,用MTT比色法繪制細(xì)胞生長(zhǎng)曲線,并用特定誘導(dǎo)液將分離的BMSCs向成骨細(xì)胞和成脂肪細(xì)胞定向誘導(dǎo)分化,利用ALP和油紅0進(jìn)行染色鑒定。結(jié)果所分離的BMSCs細(xì)胞在形態(tài)學(xué)觀察與生長(zhǎng)動(dòng)力學(xué)上均符合B
2、MSCs特征,分離培養(yǎng)的BMSCs細(xì)胞在第3天進(jìn)入對(duì)數(shù)生長(zhǎng)期,第10天進(jìn)入平臺(tái)期;在成骨、成脂肪的誘導(dǎo)培養(yǎng)條件下,分別出現(xiàn)成骨、成脂肪表型特征,可進(jìn)一步定向分化,結(jié)論所收獲的細(xì)胞具有BMSCs的特異性。【關(guān)鍵詞】骨髓間充質(zhì)干細(xì)胞;分離;成骨分化;成脂分化[Abstract】ObjectiveToestablishasustainedandstablebonemarrowderivedmesenchymalstemcells(BMSCs)isolationandculturesystemwhichcouldbeplur

7、cdifferentiation;Adipogenicdifferentiation軟骨組織工程技術(shù)(自體軟骨細(xì)胞的移植、生長(zhǎng)因子對(duì)軟骨細(xì)胞的作用、骨髓間充質(zhì)干細(xì)胞(又稱骨髓基質(zhì)干細(xì)胞,BMSCs)在軟骨組織工程中的作用、轉(zhuǎn)基因技術(shù)〕是當(dāng)今的研究熱點(diǎn)(1,2)0特別是自體BMSCs,因其低免疫原性、無(wú)限擴(kuò)增性及多向分化潛能等諸多優(yōu)點(diǎn)而引起廣泛關(guān)注,成為軟骨組織工程種子細(xì)胞的首選來(lái)源。本實(shí)驗(yàn)選擇合適的BMSCs體外分離、純化和擴(kuò)增方法,并通過(guò)形態(tài)學(xué)觀察(光鏡、電鏡)、細(xì)胞周期(流式細(xì)胞儀)、多細(xì)胞系(成骨細(xì)胞,脂肪細(xì)胞
