1、論著里氏木霉產(chǎn)胞外多糖發(fā)酵條件的優(yōu)選研究**陳鑫劉小燕閔敏田銳黃政政文志田曄(安徽理工大學(xué)醫(yī)學(xué)院,安徽淮南232001)【摘要】目的建立簡便有效的里氏木霉胞外多糖提取發(fā)酵工藝,為木霉胞外多糖的進(jìn)一步開發(fā)利用奠定基礎(chǔ)。方法以粗胞外多糖產(chǎn)量為指標(biāo),采用單因素試驗(yàn)優(yōu)化發(fā)酵培養(yǎng)基配方,利用正交試驗(yàn)對搖瓶培養(yǎng)過程中的發(fā)酵液初始pH、孢子接種量、發(fā)酵溫度及發(fā)酵時(shí)間進(jìn)行優(yōu)選。結(jié)果里氏木霉發(fā)酵最佳控制參數(shù)為:培養(yǎng)基配方為蔗糖50g/L、酵母粉20g/L、K2HPO·43H2O8g/L、MgSO40.5g/L;發(fā)酵條件為發(fā)酵液初始pH5.0、孢子接種量10%、發(fā)酵溫度28℃、發(fā)酵時(shí)間

7、步研究木霉胞外多糖的生物活性奠定實(shí)驗(yàn)條件簡單,分布廣泛,環(huán)境適應(yīng)性強(qiáng),營養(yǎng)生長迅速,且生長代基礎(chǔ),亦為木霉胞外多糖的大規(guī)模生產(chǎn)應(yīng)用提供基礎(chǔ)研究依據(jù)。謝過程中可分泌大量活性胞外多糖。胞外多糖1材料與方法(exopolysaccharide,EPS)指微生物在生長代謝過程中分泌到細(xì)1.1材料胞外的一類多糖,真菌EPS具有豐富的生物學(xué)功能,可發(fā)揮抗1.1.1菌種實(shí)驗(yàn)用里氏木霉菌株T1由本實(shí)驗(yàn)室分離保腫瘤、免疫調(diào)節(jié)、抗炎癥、抗菌、抗氧化及益生等多種功能[2]。研存。究發(fā)現(xiàn),木霉胞外多糖亦具有抗菌、抗腫瘤和免疫增強(qiáng)等多種1.1.2培養(yǎng)基配方液體發(fā)酵基礎(chǔ)培養(yǎng)基:葡萄糖50g
