4、vedevelopedamodelsystemtostudytherolesofboththehostandtheparasitebyusingnude(nu/nu)miceandatemperature-sensitivemutantofvesicularstomatitisvirus(tsG31VSV).Whenthenudemicewereintracerebrallyinfectedwiththetemperature-sensitiveVSV,allanimalsdiedafter12daysofinfection.If,howeve
5、r,themicewererecon-stitutedwithsyngeneicT-lymphocytes,70%oftheanimalssurvivedforatleast21days.TheamountofantibodyagainstthevirusproducedinthemicewasdependentuponhowmanyT-lymphocyteswereusedtoreconstitutetheanimals.InfectiousVSVwasisolatedfromthebrainofalltheinfectedanimalsth
6、atdidnothavehightitersofantibodyintheirsera.WhenthebrainisolatedVSVwasinoculatedintoSwissoutbredmice,mostoftheanimalsdied,incontrasttotsG31VSVwherelessthan5%oftheanimalssuccumbed.Tofurthercharacterizethebrainisolatedvirus,theviruswasclonedandanalyzedfortemperature-sensitivit
7、y.Allthecloneswerelesstemperature-sensitivethantheoriginaltsG31VSV,andmostoftheclonescausedamoreaggressivediseasewheninfectedintoSwissoutbredmice.UnlikethetsG31VSV,mostoftheclonalisolateshadatemperature-sensitivedefectinRNAsynthesis.StudiesarecurrentlyunderwaytolearnhowT-lym
