1、·基礎(chǔ)論著·大鼠腦皮質(zhì)未成熟星形膠質(zhì)細(xì)胞的分離、鑒定及原代培養(yǎng)吳炎宇侯博王伯丞郭英【摘要】目的探討大鼠腦皮質(zhì)未成熟星形膠質(zhì)細(xì)胞的分離、鑒定及原代培養(yǎng)技術(shù)。方法采用差速貼壁法清除成纖維細(xì)胞及酶消化去除少突膠質(zhì)細(xì)胞污染,應(yīng)用免疫熒光進(jìn)行細(xì)胞鑒定。取不同時(shí)間點(diǎn)的細(xì)胞在倒置顯微鏡下觀察細(xì)胞形態(tài)變化,雙重免疫熒光染色觀察細(xì)胞生長(zhǎng)過(guò)程的分化狀態(tài)。結(jié)果細(xì)胞形態(tài)觀察提示:6~14d細(xì)胞呈均勻扁平多角狀,16d后部分細(xì)胞肥大增生,呈成熟狀態(tài)。免疫組化染色顯示:GFAP(+)細(xì)胞達(dá)97%。免疫熒光雙重標(biāo)記發(fā)現(xiàn):16d后細(xì)胞表達(dá)vimentin和nes

6、eastrocytescellsfromratbraincortex.[Keywords]Glialfibrillaryacidicprotein;Vimentin;Nestin;Immatureastrocytes未成熟星形膠質(zhì)細(xì)胞作為星形膠質(zhì)細(xì)胞的前的修復(fù),成為了當(dāng)今研究神經(jīng)修復(fù)的熱點(diǎn)[3】。然而體細(xì)胞具有定向分化成成熟星形膠質(zhì)細(xì)胞的潛能,對(duì)于如何快速獲取大量純化后的未成熟星形膠質(zhì)但相對(duì)于成熟星形膠質(zhì)細(xì)胞而言更具有可塑性。研細(xì)胞是神經(jīng)修復(fù)研究的關(guān)鍵內(nèi)容。隨著細(xì)胞學(xué)研究究表明,未成熟的星形膠質(zhì)細(xì)胞分泌一類蛋白促進(jìn)的發(fā)展,目前對(duì)于
