2、于六孔板,分為3組:以E2F.1沉默重組慢病毒顆粒(Lv—shRNA.E2F.1)感染SGC-7901/DDP,作為L(zhǎng)v.shRNA.E2F.1組;以陰性對(duì)照慢病毒顆粒(Lv.shRNA-NC)感染SGC一7901/DDP,作為L(zhǎng)v-shRNA-NC組;空白對(duì)照組不做任何處理。以MTT法測(cè)定轉(zhuǎn)染后各組細(xì)胞對(duì)DDP的半數(shù)抑制濃度(Ic。),并以AxnnexinV-PE/7-AAD雙染色流式細(xì)胞術(shù)檢測(cè)各組細(xì)胞凋亡率及周期分布。應(yīng)用Westernblot和半定量RT-PCR技術(shù)分別對(duì)各組細(xì)胞中E2F-1、survivin、Bc1.2的mRNA及蛋白水
3、平進(jìn)行檢測(cè)。結(jié)果與Lv.shRNA.NC組和空白對(duì)照組相比,Lv—shRNA-E2F一1組細(xì)胞對(duì)順鉑的Ic。顯著降低,細(xì)胞凋亡率顯著提高,細(xì)胞周期阻滯于GdG期,差別均有統(tǒng)計(jì)學(xué)意義(尸<0.05);Lv·shRNA-E2F-1組E2F.1mRNA水平較Lv.shRNA.NC組和空白對(duì)照組分別下降45.0%和41.3%,蛋白水平分別下降66.7%和70.5%(均P<0.01);Lv-shRNA-E2F一1組survivinmRNA水平較Lv.shRNA.NC組和空白對(duì)照組分別下降30.3%和28.7%,蛋白水平分別下降56.5%和53.6%(均P
4、<0.01);Lv.shRNA.E2F.1組Bcl一2mRNA水平較Lv—shRNA.NC組和空白對(duì)照組分別下降76.6%和76.8%,蛋白水平分別下降74.6%和79.9%(均P<0.01);Lv.shRNA.NC組與空白對(duì)照組比較,上述指標(biāo)差別均無統(tǒng)計(jì)學(xué)意義(均P>0.05)。結(jié)論:沉默E2F.1基因可有效提高人胃癌耐藥細(xì)胞株SGC.7901/DDP對(duì)順鉑的敏感性,其機(jī)制可能與survivin及Bcl一2表達(dá)下調(diào)有關(guān),提示E2F.1可能成為胃癌藥物治療的新靶點(diǎn)。關(guān)鍵詞:E2F一1;胃癌;cisplatin;化療敏感性;survivin;Bc
