1、蚌埠醫(yī)學院學報2014年4月第39卷第4期433[文章編號]1000-2200(2014)04-0433-05·基礎(chǔ)醫(yī)學·I3整合素反義寡核苷酸聯(lián)合順鉑治療人卵巢癌裸鼠移植瘤的研究馬玲，田敏[摘要]目的：探討以13整合素反義寡核苷酸(ASODN)治療人卵巢癌裸鼠皮下移植瘤的可行性。方法：常規(guī)體外培養(yǎng)SKOV3細胞，采用皮下注射法建立移植瘤裸鼠動物模型。32只荷瘤鼠隨機分為ASODN組(A組)、ASODN聯(lián)合順鉑(DDP)組(A+D組)、DDP組和0．9％氯化鈉注射液對照組(Ns組)，每組8只，分組給藥。以脂質(zhì)體包裹的母整合素ASODN直接移植瘤內(nèi)注射，觀察腫
2、瘤生長情況，測瘤體積并計算抑瘤率。采用反轉(zhuǎn)錄一聚合酶鏈反應(yīng)(RT-PCR)和免疫組織化學方法分別檢測p整合素mRNA的表達。結(jié)果：A組腫瘤體積和抑瘤率分別為(316．10±21．77)mm和48．15％，與Ns組比較抑瘤率較高，腫瘤生長緩慢(P<0．01)。而A+D組腫瘤體積和抑瘤率分別為(178．70±40．67)mm和7O．37％，與DDP組、A組及NS組差異均有統(tǒng)計學意義(P<0．05～P<0．01)。RT-PCR和免疫組織化學檢測，A組和A+D組腫瘤組織中B整合素mRNA的表達均明顯下調(diào)(P<0．01)。結(jié)論：單用B。整合素ASDON或聯(lián)用DDP均可
