1、山東醫(yī)藥2014年第54卷第2O期阿霉素對(duì)大腸癌SW480細(xì)胞周期及周期相關(guān)蛋白表達(dá)的影響孟永亮。孫彥(1山東萬(wàn)杰醫(yī)學(xué)院，山東淄博255213；2山東省高校生物醫(yī)學(xué)工程技術(shù)重點(diǎn)實(shí)驗(yàn)室)摘要：目的探討阿霉素對(duì)大腸癌SW480細(xì)胞周期及周期相關(guān)蛋白表達(dá)的影響。方法體外培養(yǎng)SW480細(xì)胞，隨機(jī)分為A(空白對(duì)照組)、B、C、D、E組，阿霉素濃度分別為0、2．5、5、10、20Ixg／mL。采用MTF法測(cè)定各組細(xì)胞培養(yǎng)l2、24、48、72h后的細(xì)胞生長(zhǎng)抑制率；流式細(xì)胞術(shù)分析各組細(xì)胞培養(yǎng)48h后細(xì)胞周期；Westernblot法測(cè)定各組細(xì)
2、胞培養(yǎng)48h后SW480細(xì)胞周期相關(guān)蛋白CyclinB1、CyclinA表達(dá)水平。結(jié)果阿霉素抑制SW480細(xì)胞增殖具有濃度和時(shí)間依賴性。不同濃度阿霉素作用于SW480細(xì)胞48h后，D、E組G。／G。期細(xì)胞百分率低于A、B、C組，SubG期細(xì)胞百分率、凋亡指數(shù)(AI)高于A、B、C組，P均<0．05；與A組比較，B、C組CyclinB1表達(dá)量增高(P均<0．05)，D、E組表達(dá)量下降(P均<0．05)；隨著阿霉素濃度增加，CyclinA的表達(dá)量逐漸降低(P<0．05)。結(jié)論阿霉素可將SW480細(xì)胞周期阻滯于Go／G．期，影響細(xì)胞周
