1、實(shí)用藥物與臨床2014年第17卷第10期PracticalPharmacyAndClinicalRemedies．2014．Vo1．17，No．10·1247·金雀異黃素對(duì)CIA大鼠促炎和抗炎因子分泌的影響馬瑩瑩，張育，許金鑫，王燕茹[摘要]目的研究金雀異黃素對(duì)Ⅱ型膠原誘導(dǎo)型關(guān)節(jié)炎大鼠促炎和抗炎因子分泌的影響。方法建立Ⅱ型膠原誘導(dǎo)型關(guān)節(jié)炎大鼠模型，通過50、100和200m的金雀異黃素治療，以0．1mg／mL每周甲氨蝶呤(MTX)作為陽性對(duì)照，治療后每周檢測(cè)1I型膠原誘導(dǎo)型關(guān)節(jié)炎大鼠關(guān)節(jié)炎指數(shù)評(píng)分和后肢容積，
2、第5周取膝關(guān)節(jié)進(jìn)行關(guān)節(jié)滑膜組織病理檢測(cè)；取靜脈血分離出血清后，用酶聯(lián)免疫吸附試驗(yàn)檢測(cè)各組大鼠血清中白細(xì)胞介素_4(IL_4)、IL-6、IL一8、IL一10、轉(zhuǎn)化生長(zhǎng)因子一B1(TGF一131)、淋巴毒素一的水平。結(jié)果關(guān)節(jié)炎指數(shù)評(píng)分、后肢容積以及關(guān)節(jié)滑膜組織病理結(jié)果顯示造模成功。模型組大鼠血清中的炎癥因子IL6、IL一8和淋巴毒素一OL的表達(dá)水平明顯高于空白對(duì)照組(P<0．05)；金雀異黃素能下調(diào)IL-6、IL一8、淋巴毒素一的分泌，200m金雀異黃素對(duì)IL-6、1L一8的抑制作用與MTX相當(dāng)，而各金雀異黃素
3、治療組對(duì)淋巴毒素一的抑制作用均弱于MTX。模型組大鼠血清中的細(xì)胞因子IL4、IL一10、TGF一131的表達(dá)水平低于空白對(duì)照組(P<0．05)；金雀異黃素能上調(diào)IL-4、IL一10、TGF一131的分泌。100m金雀并黃素對(duì)IL4的上調(diào)作用與MTX相當(dāng)；200m金雀異黃素對(duì)IL一10的促進(jìn)作用與MTX相當(dāng)，對(duì)TGF一131的上調(diào)作用強(qiáng)于MTX。結(jié)論金雀異黃素抑制Ⅱ型膠原誘導(dǎo)型關(guān)節(jié)炎大鼠炎癥因子的釋放，促進(jìn)抗炎因子的表達(dá)，改善關(guān)節(jié)炎大鼠滑膜炎癥。[關(guān)鍵詞]金雀異黃素；甲氨蝶呤；CIA；細(xì)胞因子Efectofge
