1、牛雙芽巴貝斯蟲(chóng)rap-1基因的克隆與序列分析摘要:對(duì)牛雙芽巴貝斯蟲(chóng)(babesiabigemina)潛在藥物靶標(biāo)rap-1的基因進(jìn)行克隆與序列分析。以蘭州株牛雙芽巴貝斯蟲(chóng)基因組為模板,經(jīng)pcr擴(kuò)增獲得了rap-1基因,將該基因克隆到pgem-teasyvector上,對(duì)重組質(zhì)粒進(jìn)行pcr擴(kuò)增,對(duì)目的基因進(jìn)行酶切鑒定及序列測(cè)定分析。結(jié)果表明,rap-1基因長(zhǎng)度為846bp,同源性分析結(jié)果顯示,克隆序列與genbank收錄的巴西株牛雙芽巴貝斯蟲(chóng)的核苷酸序列同源性為99.74%,說(shuō)明蘭州株牛雙芽巴貝斯蟲(chóng)的rap-1與genbank公布的參考基

8、白尿等,可引起宿主死亡[2],該病是世界公認(rèn)的危害養(yǎng)牛業(yè)的主要寄生蟲(chóng)病之一。牛雙芽巴貝斯蟲(chóng)入侵紅細(xì)胞的過(guò)程是感染發(fā)病的重要環(huán)節(jié),對(duì)紅細(xì)胞的入侵是由位于牛雙芽巴貝斯蟲(chóng)細(xì)胞膜表面的頂體復(fù)合器以及棒狀體上的相關(guān)蛋
