1、2013—年第16卷第12期PracticalPharmacyAndClinicalRemedies,2013Vo1.16.No.12·論著·丙戊酸聯合順鉑對人肝癌細胞的增殖抑制作用研究蔣鳳琴,袁玉梅,汪榮華,姚沖,曹恒斌[摘要]目的探討丙戊酸(VPA)聯合順鉑(DDP)對人肝癌細胞HepG2、BEC7404、SMMC7721的增殖抑制作用。方法三組濃度的VPA與DDP單藥及聯合作用于人肝癌細胞HepG2、BEC7404、SMMC7721.24、48、72h后,觀察細胞數量和形態(tài)的變化。用MTT法分析藥物對細胞增殖的抑制率(IR),
2、計算聯合用藥對細胞增殖抑制率q值,考察聯合使用是否為協(xié)同作用。結果72h后,各用藥組細胞數目減少十分明顯,其中vPA+DDP組較單獨用藥組減少更為顯著,其細胞形態(tài)發(fā)生較大改變。采用MTT比色法顯示,對照組細胞生長率明顯高于各實驗組,聯合用藥組對細胞增殖IR更為顯著,且隨著藥物濃度增加及作用時間延長,各組對細胞增殖IR明顯增加。根據q值結果,在VPA較高濃度(≥100g/mE)時,聯合用藥組具有明顯的協(xié)同作用。結論VPA能增強DDP對人肝癌細胞的增殖抑制作用,并且在VPA較高濃度時,兩藥具有協(xié)同優(yōu)勢。[關鍵詞]丙戊酸;順鉑;肝癌細胞;
