2、從新生大鼠胰島中分離培養(yǎng)胰島祖細(xì)胞,用含20nmol/LGLP一1(7—36)NH2的RPMI1640完全培養(yǎng)基誘導(dǎo)分化4周(GLP-1組,n=5),以RP—MI1640完全培養(yǎng)基作對照(n=5).免疫細(xì)胞化學(xué)染色檢測誘導(dǎo)分化前后細(xì)胞Nestin,PDX-1,胰島素,生長抑素的表達(dá);放射免疫法測定每周末培養(yǎng)基中胰島素含量(胰島素分泌量);二硫腙染色觀察形成的胰島樣細(xì)胞團(tuán)(ICCs),4周后計數(shù)每25am中直徑>50}xm的ICCs數(shù)目.結(jié)果:誘導(dǎo)前胰島祖細(xì)胞不表達(dá)PDX-1,胰島素,生長抑素.僅表達(dá)Nestin.誘導(dǎo)4周后,GLP一1組PDX一1,胰島素,生長抑素表達(dá)陽
