2、的食管癌細(xì)胞Eca109(DDPr)。冰片聯(lián)合DDP作用于Eca109(DDPr)細(xì)胞,MTT法檢測Eca109(DDPr)細(xì)胞增殖情況;流式細(xì)胞術(shù)檢測細(xì)胞周期及細(xì)胞凋亡。結(jié)果:遞增藥物質(zhì)量濃度持續(xù)作用誘導(dǎo)6個(gè)月后,細(xì)胞可以在含1μg/mlDDP的培養(yǎng)液中正常生長,其對DDP的耐藥指數(shù)為15.886,所得細(xì)胞命名為耐DDP的食管癌細(xì)胞Eca109(DDPr)。單用冰片濃度≤1.560μg/ml時(shí),對Eca109(DDPr)細(xì)胞無明顯抑制作用(P>0.05),但與DDP1μg/ml合用能顯著促進(jìn)DDP抑制Eca109(DDPr)細(xì)胞
3、的增殖(P<0.05),并促進(jìn)DDP誘導(dǎo)Eca109(DDPr)細(xì)胞周期改變、細(xì)胞凋亡增加。細(xì)胞周期表現(xiàn)為G1期細(xì)胞明顯增多(P<0.05),S期細(xì)胞、G2期細(xì)胞明顯減少(P<0.05);細(xì)胞的凋亡率明顯增加(P<0.05)。結(jié)論:冰片促進(jìn)DDP抑制Eca109(DDPr)細(xì)胞增殖可能與改變Eca109(DDPr)細(xì)胞周期、增加細(xì)胞凋亡有關(guān)。[關(guān)鍵詞]冰片;順鉑;Eca109(DDPr);細(xì)胞增殖;細(xì)胞周期;細(xì)胞凋亡[中圖分類號(hào)]R735.1[文獻(xiàn)標(biāo)識(shí)碼]AEffectsofBorneolcombinedwithcisplatin
