1、JNK在驚厥持續(xù)狀態(tài)幼年大鼠海馬中的表達(dá)及依達(dá)拉奉對(duì)其的影響作者:陸海燕,鄧小龍,李光乾,李偉【摘要】目的:研究(JNK)在驚厥持續(xù)狀態(tài)(SC)幼年大鼠海馬中的表達(dá)及依達(dá)拉奉對(duì)其的影響。方法:雄性SD幼年大鼠120只分為正常對(duì)照組(NC組)、驚厥持續(xù)狀態(tài)組(SC組)、依達(dá)拉奉組(EDA組)三大組;各大組均隨機(jī)分為5組,分別于驚厥后4、12、24、48和72h處死。采用氯化鋰-匹魯卡品法制作驚厥持續(xù)狀態(tài)模型。EDA組在大鼠造模前三天予EDA腹腔注射,每天注射一次,連續(xù)3d。TUNEL法檢測(cè)觀察大鼠海馬細(xì)胞凋亡情況;免疫組化法檢測(cè)海馬p-JNK蛋白表達(dá)動(dòng)態(tài)變化;RT-PCR技術(shù)檢測(cè)
2、海馬JNK1mRNA表達(dá)的動(dòng)態(tài)變化。結(jié)果:SC組在驚厥12h后的各時(shí)間點(diǎn)海馬CA1區(qū)TUNEL陽(yáng)性細(xì)胞數(shù)均顯著高于NC組(P<0.01),而ED組TUNEL陽(yáng)性細(xì)胞數(shù)均較SC組顯著下降(P<0.01或P<0.05)。幼年大鼠海馬CA1區(qū)p-JNK蛋白在SC組表達(dá)顯著增強(qiáng),與NC組比較差異有顯著性(P<0.01),在EDA組表達(dá)較SC組明顯下降(P<0.01)。SC組海馬JNK1mRNA表達(dá)明顯高于NC組(P<0.01),而在EDA組JNK1mRNA的表達(dá)較SC組顯著降低(P<0.01)。結(jié)論:SC可誘導(dǎo)JNK的表達(dá),促進(jìn)SC后海馬神經(jīng)元

7、ndEDAgroupwasmuchlowerthanSCgroup(P<0.01).TheexpressionofJNK1mRNAinthehippocampusinSCGroupwasmuchhigherthanNCGroup(P<0.01),andEDAgroupwassignificantlylowerthanSCgroup(P<0.01).Conclusion:SCmaybeincreasetheexpressionofJNK,JNKmaybemediatedtoc
