1、依達(dá)拉奉對幼鼠驚厥持續(xù)狀態(tài)后海馬XBP1mRNA和caspase12表達(dá)及神經(jīng)元凋亡的影響作者:鄧小龍,王海萍,焦穎,林忠東,李光乾【摘要】目的:探討依達(dá)拉奉對幼鼠驚厥持續(xù)狀態(tài)(statusconvulsion,SC)后海馬內(nèi)質(zhì)網(wǎng)應(yīng)激(endoplasmicreticulumstress,ERS)關(guān)鍵標(biāo)志分子X盒結(jié)合蛋白1(X-boxbindingprotein1,XBP1)mRNA、caspase-12的表達(dá)及神經(jīng)元凋亡的影響。方法:將19~21日齡SD大鼠隨機(jī)分入驚厥持續(xù)狀態(tài)(SC)組、依達(dá)拉奉(ED)組、生理鹽水對照(NS)組;各組再按不同時間點(diǎn)分五個亞組。應(yīng)用氯化鋰-匹魯卡
2、品建立大鼠SC模型,用半定量逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)動態(tài)觀察SC后海馬XBP1、caspase-12mRNA的表達(dá)情況。用免疫組織化學(xué)方法觀察海馬caspase-12蛋白表達(dá)情況。用TUNEL法觀察神經(jīng)元凋亡細(xì)胞數(shù)。結(jié)果:幼年大鼠海馬中XBP1和caspase-12在SC組表達(dá)顯著增強(qiáng),與NS組比較差異有顯著性(P<0.01);與SC組比較,ED組XBP1mRNA表達(dá)顯著升高(P<0.01或P<0.05),caspase-12mRNA及蛋白表達(dá)顯著降低(P<0.01或P<0.05);SC組在驚厥1215h后的各時間點(diǎn)海馬CA1區(qū)TUNEL陽
