1、survivin反義寡核苷酸誘導(dǎo)胃癌原代細(xì)胞凋亡的實(shí)驗(yàn)研究【摘要】目的探討survivin反義寡核苷酸對(duì)人胃癌原代細(xì)胞凋亡的影響,為胃癌的靶向治療提供實(shí)驗(yàn)依據(jù)。方法利用人胃癌實(shí)體瘤新鮮標(biāo)本培養(yǎng)胃癌原代細(xì)胞,胃癌細(xì)胞分為空白對(duì)照組、脂質(zhì)體組、正義寡核苷酸組及反義寡核苷酸轉(zhuǎn)染組。用脂質(zhì)體介導(dǎo)survivin反義寡核苷酸轉(zhuǎn)染胃癌原代細(xì)胞,48h后,觀察腫瘤細(xì)胞的形態(tài)變化,Westernblot檢測(cè)survivin蛋白的表達(dá),流式細(xì)胞術(shù)檢測(cè)細(xì)胞的凋亡。結(jié)果反義寡核苷酸轉(zhuǎn)染組腫瘤細(xì)胞體積變小,細(xì)胞碎裂,出現(xiàn)凋亡小體;survi
2、vin蛋白量下降;流式細(xì)胞術(shù)檢測(cè)到腫瘤細(xì)胞的凋亡率增高。與其他各組相比,其差異有統(tǒng)計(jì)學(xué)意義(P<0.05),而空白對(duì)照組、脂質(zhì)體組、正義寡核苷酸組之間的差異無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論survivin反義寡核苷酸在脂質(zhì)體介導(dǎo)下轉(zhuǎn)染人胃癌原代細(xì)胞,可以誘導(dǎo)腫瘤細(xì)胞發(fā)生凋亡,抑制細(xì)胞生長(zhǎng),反義survivin可能成為一個(gè)促進(jìn)胃癌細(xì)胞凋亡的手段?!娟P(guān)鍵詞】survivin;反義寡核苷酸;凋亡;原代細(xì)胞;胃癌Researchoneffectsofsurvivinantisenseoligonucleotid
