1、survivin反義寡核苷酸激活caspase-3誘導(dǎo)SGC7901胃癌細(xì)胞凋亡的實(shí)驗(yàn)研究【摘要】目的研究轉(zhuǎn)染生存素(survivin)反義寡核苷酸誘導(dǎo)SGC7901胃癌細(xì)胞凋亡及其可能的作用機(jī)制。方法實(shí)驗(yàn)分為空白對照組(Control組)、脂質(zhì)體組(Lip組)、正義寡核苷酸組(Sodn組)及反義寡核苷酸組(Asodn組)。人工合成正、反義寡核苷酸,經(jīng)脂質(zhì)體將survivin正、反義寡核苷酸轉(zhuǎn)染入胃癌細(xì)胞48h后,MTT檢測細(xì)胞增殖抑制率,Westernblot檢測caspase-3蛋白表達(dá)改變,Hoechst染色觀察細(xì)胞凋亡形態(tài)
2、改變及流式細(xì)胞儀檢測凋亡率。結(jié)果Hoechst染色熒光顯微鏡下Control組、Lip組及Sodn組細(xì)胞核呈正常的藍(lán)色,而Asodn組細(xì)胞核染色增強(qiáng)、核質(zhì)濃縮、核碎裂,呈凋亡改變;Asodn組細(xì)胞增殖抑制率、細(xì)胞凋亡率、caspase-3蛋白表達(dá)均增高,與Control組、Lip組及Sodn組相比差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論survivin反義寡核苷酸可誘導(dǎo)胃癌細(xì)胞凋亡,其作用機(jī)制可能是通過激活caspase-3表達(dá),啟動凋亡信號傳導(dǎo)?!娟P(guān)鍵詞】生存素;反義寡核苷酸;caspase-3;胃癌細(xì)胞;凋亡Abstra

4、sodn).Artificiallysynthesizedsurvivinantisenseoligonucleotidewastransfectedfor48hoursintogastriccancercellsusingliposome.Aftercultivation,thecellproliferationinhibitionratewasevaluatedbyMTTassay,theexpressionofcaspase-3proteinwasdeterminedwithWesternblot,andthemorphol
5、ogicalchangesofapoptosiswereobservedbyHoechststaining,andtheapoptosisratewastestedwithAnnexinv-FITCassayofflowcytometricanalysis.ResultsFluorescencemicroscopicobservationsrevealedthatHoechststaininginthecontrol,LipandSodngroupshadnucleuspresentedasnormalblue,whiletheA
6、sodngrouphadsuchchangesasreinforcednucleusstaining,karyoplasmconcentration,karyorrhexisandapoptoticmodifications.Asodngrouphadasignificantincreaseinthecellproliferationinhibitionrate,cellapoptosisrateandexpressionofcaspase-3protein,comparedwiththecontrol,LipandSodngro
