1、長春工業(yè)大學(xué)人文信息學(xué)院學(xué)士學(xué)位論文摘要目的：人參蛋白及多肽主要具有抗脂質(zhì)分解、抗血紅細(xì)胞聚積、抗真菌、抗病毒、精氨酸酶活性等。選擇研究較少但活性多樣的大分子－人參蛋白及多肽，通過現(xiàn)代分離純化技術(shù)、活性檢測(cè)技術(shù)及分析鑒定技術(shù)進(jìn)行生物特性研究。通過對(duì)不同參之間的蛋白成分進(jìn)行比較和研究，對(duì)人參的一些多肽成分的分析和應(yīng)用提供依據(jù)。方法：采用中藥化學(xué)及生物化學(xué)提取法,結(jié)合離子交換、凝膠過濾層析和反相C18層析等現(xiàn)代生物分離技術(shù),利用膜分離技術(shù)及柱層析技術(shù)對(duì)總蛋白進(jìn)行分離純化，結(jié)合細(xì)胞活性實(shí)驗(yàn)進(jìn)行活性篩選，對(duì)人參中的蛋白及多肽進(jìn)
2、行了分離純化，采用HPLC、SDS-聚丙烯酰胺凝膠電泳法（SDS-PAGE）及凝膠電泳圖像分析軟件等譜學(xué)技術(shù)，初步研究了這些蛋白多膚的結(jié)構(gòu),并從細(xì)胞水平進(jìn)行了體外活性實(shí)驗(yàn),探討蛋白與多膚的生物活性。結(jié)果：采用現(xiàn)代生物層析分離技術(shù)從人參中分離純化得到3個(gè)蛋白質(zhì)單體化合物,分別為GSPⅠ、GSPⅡ及GSPⅢ。經(jīng)基質(zhì)輔助激光解吸電離飛行時(shí)間質(zhì)譜人參蛋白及多肽藥理活性實(shí)驗(yàn)表明,人參蛋白GSPll對(duì)金黃色葡萄球菌具有抑制作用;對(duì)Hep-2細(xì)胞具有抑制作用，對(duì)3T3細(xì)胞具有增殖作用，且活性呈現(xiàn)一定量效關(guān)系。關(guān)鍵詞人參多肽提取工藝活
