1、單向灌流法研究巴戟天低聚糖的大鼠腸吸收特性 [摘要]采用大鼠在體腸單向灌流實(shí)驗(yàn)?zāi)P?,利用HPLC-ELSD測(cè)定腸灌流液中蔗糖、蔗果三糖、耐斯糖、1F-果呋喃糖基耐斯糖和巴戟甲素等5種低聚糖的含量,研究各低聚糖在小腸段(十二指腸、空腸、回腸)和結(jié)腸的吸收情況,并考察藥物濃度、灌流液pH以及P-糖蛋白(P-gp)抑制劑對(duì)藥物吸收的影響,得出各低聚糖成分在大鼠體內(nèi)的腸吸收機(jī)制。結(jié)果顯示5種低聚糖均為全腸道吸收的藥物,吸收速率與灌流液pH、藥物濃度和腸段部位有關(guān)。鹽酸維拉帕米可顯著增加蔗糖與巴戟甲素的吸收量,提示蔗糖與巴戟甲素是P-gp的底物。5種低聚糖在大鼠腸道內(nèi)的吸收機(jī)制以被動(dòng)擴(kuò)散方
