1、中文摘要。目的用化學(xué)方法去除日本大耳白兔雙側(cè)脛神經(jīng)的雪旺氏細(xì)胞及髓鞘,制成去細(xì)胞神經(jīng)基膜管,將NGF(神經(jīng)生長因子)與神經(jīng)基膜管復(fù)合,橋接成年wiStar大鼠坐骨神經(jīng)10mm神經(jīng)缺損,進(jìn)一步探索NGF與異種去細(xì)胞神經(jīng)基膜管復(fù)合后用于異種移植修復(fù)周圍神經(jīng)缺損的效果。方法選用成年日本大耳白兔15只,切取兔雙側(cè)脛神經(jīng)各4一,隨機抽取15例新鮮神經(jīng),分別取其中2∞行流式細(xì)胞法檢測M}IC一Ⅱ類組織相容性抗原,其余2crn行去細(xì)胞處理后再行流式細(xì)胞法檢測MHC一Ⅱ類抗原及組織學(xué)、電鏡觀察其超微結(jié)構(gòu),另15例新鮮神經(jīng)分別剪成1—長度,經(jīng)化學(xué)方法制成去
2、細(xì)胞神經(jīng)基膜管,其中15段用4500U/“的NGF溶液4℃浸泡24小時;另15段置于無菌PBS緩沖液中4℃保存?zhèn)溆?;根?jù)移植物的不同,將wistar大鼠隨機分為3組,每組15只:實驗組(A組):去細(xì)胞異種神經(jīng)基膜管復(fù)合NGF移植組、對照組I(B組):去細(xì)胞異種神經(jīng)基膜管移植組、對照組Ⅱ(C組):自體神經(jīng)移植組。分籠喂養(yǎng),術(shù)后1個月行神經(jīng)電生理檢測,包括脛后肌群運動誘發(fā)電位,再生神經(jīng)的傳導(dǎo)速度;用HE染色,免疫組化染色,透射電鏡等方法對移植段遠(yuǎn)端吻合口再生神經(jīng)纖維進(jìn)行形態(tài)學(xué)觀察,并對再生有髓神經(jīng)纖維的數(shù)量、密度、直徑及雪旺氏細(xì)胞的密度進(jìn)行量
3、化分析。結(jié)果移植前新鮮神經(jīng)組MHc一Ⅱ類抗原檢測值為72.14±19.88,去細(xì)胞組MHC一Ⅱ類抗原檢測值為4.19±3.11,兩組比較存在顯著性差異(PO.05),A組與B組比較差異有顯著性(P<0.05),說明神經(jīng)恢復(fù)效果優(yōu)于B組。組織學(xué)觀察見3組移植體遠(yuǎn)端吻合口橫切面
