1、中國(guó)摩虧通報(bào)2009,25(07):62—66ChineseAgriculturalScienceBulletin農(nóng)桿菌介導(dǎo)莖尖轉(zhuǎn)化體系對(duì)不同基因型棉花轉(zhuǎn)化率的影響孫旭,蘇翔,凌磊,蔡沂,洪薩麗,林毅,蔡永萍(安徽農(nóng)業(yè)大學(xué)生命科學(xué)學(xué)院,合肥230036)摘要:選用兩種不同基因型陸地棉華抗6號(hào)和06S62為材料,以無(wú)菌黃化苗莖尖為受體,用農(nóng)桿菌介導(dǎo)法進(jìn)行轉(zhuǎn)化,對(duì)不同的菌液濃度、浸染時(shí)間和負(fù)壓處理等進(jìn)行了比較試驗(yàn),探討農(nóng)桿菌介導(dǎo)對(duì)兩種不同基因型棉花遺傳轉(zhuǎn)化率的影響。結(jié)果表明,農(nóng)桿菌介導(dǎo)棉花轉(zhuǎn)化的適宜條件是:對(duì)于華抗6號(hào),茵液濃度OD一達(dá)到0.7、浸染時(shí)間
2、10min、負(fù)壓處理10min時(shí),轉(zhuǎn)化效果較好;對(duì)于06S62,茵液濃度OD6oo達(dá)到0.9、浸染時(shí)間15min、負(fù)壓處理15min時(shí),轉(zhuǎn)化效果較好;華抗6號(hào)的轉(zhuǎn)化率高于06S62,其最高轉(zhuǎn)化率為8.81%。通過(guò)該莖尖遺傳轉(zhuǎn)化體系,將bar導(dǎo)入上述兩種基因型棉花獲得了抗除草劑的轉(zhuǎn)基因植株。關(guān)鍵詞:棉花;根癌農(nóng)桿菌;莖尖;轉(zhuǎn)化率;bar中圖分類號(hào):O37;$562文獻(xiàn)標(biāo)識(shí)碼:A。IlheEfectofAgrobac~um—mediatedTransformationSystemofShootApexonTransformationFrequencyof

7、花是中國(guó)重要的經(jīng)濟(jì)作物,常年栽培面積在耗費(fèi)大量的人力和物力。在當(dāng)前農(nóng)業(yè)結(jié)構(gòu)調(diào)整中,棉530余萬(wàn)hm,有資料表明,雜草造成直接經(jīng)濟(jì)損失占花生產(chǎn)因用工量大而處于不利地位。培育抗除草劑棉作物總產(chǎn)值的10%~20%左右,在美國(guó),平均每年因雜花,可以減少用工,降低植棉成本,對(duì)提升中國(guó)棉花育草減產(chǎn)10%,折合12億美元。在中國(guó),棉田除草需要種水平,增加棉田經(jīng)濟(jì)效益,具有重要意義。基金項(xiàng)目:安徽省“十一五”科技攻關(guān)項(xiàng)目(07010302133);安徽省教育廳重點(diǎn)項(xiàng)目(KJ2008A044)資助。第一作者簡(jiǎn)介:孫旭,女,1984年出生,安徽合肥人,碩士,從事植物生物
