1、摘要鈣是植物應(yīng)答多種生物和非生物逆境脅迫的重要信使。胞質(zhì)自由鈣濃度的變化通過對下游鈣調(diào)節(jié)蛋白的活性調(diào)控而進行信息傳遞,并最終調(diào)節(jié)相關(guān)的生理生化過程,這是植物適應(yīng)環(huán)境變化和逆境的主要機制之一。鈣依賴的蛋白激酶(CDP融)作為植物特有的一類鈣受體蛋白激酶參與并調(diào)控了許多生理過程和逆境反應(yīng)。研究表明,某些CDPKs也參與了植物響應(yīng)病原微生物和非生物脅迫的信號轉(zhuǎn)導(dǎo)途徑,但其具體功能尚不明確。本論文以模式植物擬南芥野生型、AtCDPKl突變體和組成型激活的轉(zhuǎn)基因植物為材料,探討了擬南芥么妃DPKZ在植物響
2、應(yīng)病原微生物和非生物脅迫誘導(dǎo)的信號轉(zhuǎn)導(dǎo)途徑中的可能作用,結(jié)果如下:1.利用Northernblot技術(shù)分析了AtCDPKl對丁香假單胞菌和非生物脅迫的響應(yīng),結(jié)果表明:AtCDPKl基因mRNA表達水平可被丁香假單胞菌(毒性和無毒)、非生物脅迫(低溫、干旱、鹽害)和植物激素(SA、ABA、MeJA)誘導(dǎo)增強,而高溫抑制其表達。2.純合的AtCDPKl基因T-DNA部分敲除突變體(cdpkl.J)和野生型接種無毒丁香假單胞菌后,突變體的AIGl基因和病程相關(guān)基因咫J、PR2的表達量略低于野生型。毒性
3、丁香假單胞菌侵染野生型與cdpkl.J突變體后,發(fā)現(xiàn)cdpkl.J氣孔在病原菌侵染后l小時不關(guān)閉,而野生型氣孔關(guān)閉。3.AtCDPKl組成型激活的轉(zhuǎn)基因擬南芥(K7.1)植株受到鹽脅迫后,AtCDPKI基因的表達量明顯比野生型強;并且.K7.1在種子萌發(fā)和幼苗生長過程中對高鹽脅迫的耐受能力都明顯比野生型強。根據(jù)以上實驗結(jié)果,推測擬南芥AtCDPKl在抗病和非生物脅迫介導(dǎo)的信號轉(zhuǎn)導(dǎo)中具有正向調(diào)節(jié)作用。關(guān)鍵詞:擬南芥;丁香假單胞菌;彳妃D腿,;突變體;非生物脅迫;氣孔;植物激素知識水壩@damdoc
