1、心臟雜志(ChinHeartJ)2008,20(3)·301··基礎(chǔ)研究·α2亞麻酸對高糖導(dǎo)致的人臍靜脈內(nèi)皮細胞損傷的影響11211張薇,賈國良,王四旺,韓淑芳,布倫(第四軍醫(yī)大學(xué):1.西京醫(yī)院心血管內(nèi)科,陜西西安710032,2.藥物研究所,陜西西安710032)摘要:目的觀察α2亞麻酸(ALA)對高糖導(dǎo)致的人臍靜脈內(nèi)皮細胞(HUVECs)損傷的影響,探討ALA在糖尿病血管并發(fā)癥防治中的作用。方法采用體外培養(yǎng)的第2~4代HUVECs,分為6組,即正常對照組、高糖組(HG組)及HG+ALA(ALA濃度分別為10
2、、50、100和200μmol/L)組。各組細胞于不同培養(yǎng)環(huán)境中培養(yǎng)72h后收集標本,用MTT比色法測定細胞活力,通過測定細胞上清液中一氧化氮(NO)和丙二醛(MDA)含量評估內(nèi)皮功能,末端脫氧核糖核酸酶介導(dǎo)的dUTP末端標記法(TUNEL法)和流式細胞儀法檢測細胞凋亡情況。結(jié)果較低濃度(10、50、100μmol/L)的ALA組中,細胞存活率和合成NO濃度均顯著高于高糖組(P<0105),MDA濃度和細胞凋亡率顯著低于高糖組(P<0105);尤以50μmol/L時為著。當ALA濃度增至200μmol/L時,細
