1、?論著°神經(jīng)生長因子誘導(dǎo)肝星狀細(xì)胞凋亡及其相關(guān)機(jī)制的研究舒建昌鄧延梅朱海燕呂霞何雅軍陳蓮香葉國榮【摘要】目的觀察神經(jīng)生長因子(nervegrowthfactor,NGF)對(duì)大鼠肝星狀細(xì)胞(hepaticstellatecelLHSC)凋亡的影響,并探討其可能的作用機(jī)制。方法體外培養(yǎng)大鼠肝星狀細(xì)胞株HSC-T6,將HSC-T6與100ng/mlNGFW育24h后,流式細(xì)胞術(shù)檢測(cè)細(xì)胞凋亡;細(xì)胞免疫化學(xué)法檢測(cè)HSC中凋亡相關(guān)基因P^'BclJ'CaspaseB蛋白表達(dá)的悄況;免疫熒光法檢測(cè)NGF、神經(jīng)生長因子低親和力受體p75NTR表達(dá)的悄況。結(jié)果NGF作用HSC后凋亡率較
2、對(duì)照組明顯增高[(22.36±9.51)%V8(5.88±1.36)%,P<0.05]o凋亡相關(guān)蛋白尸)、3吟?3的陽性細(xì)胞百分率與對(duì)照組比較明顯增高[(78.41±4.00)%、(39.26±1.57)%vs(34.96±3.84)%.(9.27±1.01)%,P<0.05],而Bcl?2表達(dá)實(shí)驗(yàn)組陽性細(xì)胞百分率較對(duì)照組降低[(18.12±1.38)%vs(91.53±2.98)%,P<0.05]oNGF作用HSC后NGF表達(dá)增多(6.53±1.40vs1.77±0.17,P<0.05),而p75NTR表達(dá)無明顯變化(3.52±0.36v84.24±0.38,P>0
