2、對目標(biāo)菌株進(jìn)行分子鑒定;通過測菌絲濕重來繪制其生長曲線,采用蛋白顯色法測定甲醛濃度的變化?!窘Y(jié)果】分離純化出1株甲醛耐受真菌J一5,其培養(yǎng)特征、顯微特征與曲霉屬(Aspergillus)相似;18SrDNA核酸序列同源性分析發(fā)現(xiàn),菌株J一5與曲霉屬的6個株系為同一類群,所處類群中18SrDNA基因序列兩兩之間的相似值均在76%以上。J-5菌株最大可耐受甲醛濃度為2000mg/L,可在192h內(nèi)將濃度為1560mg/L的甲醛降解到100mg/L以下。J-5菌株在培養(yǎng)0~96h內(nèi)生長緩慢,但甲醛濃度下降迅速,當(dāng)甲醛濃度下降91]400mg/L后,菌株生長
