1、生物技術(shù)通報(bào)·研究報(bào)告·8loTECHNoLOGYBULLETIN2O14年第6期不同誘導(dǎo)條件下家蠅(Muscadomestica)三齡幼蟲免疫相關(guān)基因的表達(dá)研究修江帆魏川川陳明明吳建偉(貴陽醫(yī)學(xué)院,貴陽550004)摘要:旨在研究在不同誘導(dǎo)條件下家蠅三齡幼蟲先天性免疫基因的表達(dá)情況。采用冷刺激、熱刺激及革蘭氏陰性茵、革蘭氏陽性菌和真菌注射誘導(dǎo)12h后提取家蠅三齡幼蟲總RNA。根據(jù)GenBank公布的家蠅磷酸甘油醛脫氫酶(GAPDH)基因、攻擊素(Attacin)、天蠶素(Cecropin)、防御素(Defensin)、雙翅肽(D
2、iptericin)、溶茵酶(Lysozyme)、熱休克蛋白(Heatshockprotein,HSP)及家蠅抗真菌肽(MAF1)基因序列進(jìn)行RT.PCR反應(yīng),以GAPDH基因?yàn)閮?nèi)參照,分析在不同誘導(dǎo)條件下家蠅三齡幼蟲先天性免疫基因的表達(dá)情況。結(jié)果顯示,不同條件誘導(dǎo)下,家蠅免疫相關(guān)基因表達(dá)顯現(xiàn)較大差異,微生物誘導(dǎo)比物理刺激誘導(dǎo)后家蠅免疫相關(guān)基因表達(dá)水平高;真茵、陽性菌誘導(dǎo)后家蠅免疫相關(guān)基因表達(dá)量最高,陰性菌次之;冷刺激誘導(dǎo)最低。微生物及物理刺激均能激活家蠅的免疫系統(tǒng),在不同條件刺激下,家蠅幼蟲機(jī)體免疫應(yīng)激反應(yīng)不同。關(guān)鍵詞:家蠅誘導(dǎo)
